Abstract:

Purpose: Isolating human induced pluripotent stem cells (hiPS)-derived mesenchymal progenitors as a new source of mesenchymal cells which can differentiate into different lineages like adipose and bone. Materials and Methods: After 7 days of hiPS1 culture on matrigle coated dishes, spindle like cells around colonies were removed by cell scraper. These cells that had mesenchymal like morphology was characterized after 4-6 passages. Mesenchymal cell surface markers CD73, CD90, CD105, CD29, CD44 and hematopoietic cell surface markers CD34 and CD45 were analyzed by flow cytometry and cells were differentiated to osteogenic and adipogenic lineages by defined medium. Results: flow cytometric analysis demonstrated that hiPS1 derived mesenchymal progenitors were %98.71 ± 0.14 CD44+, %98.51 ± 1.02 CD29+, %87.74 ± 3.41 CD105+, %46.65 ± 5.76 CD73+, %98.53 ± 0.78 CD90+and they did not express CD34 and CD45. These cells could be differentiated to osteogenic and adipogenic lineages. Conclusion: hiPS1 can make mesenchymal progenitors and these cells can be a suitable substitute for mesenchymal stem cells.

Fingolimod Enhances Oligodendrocyte Differentiation of Transplanted Human Induced Pluripotent Stem Cell-Derived Neural Progenitors

Multiple sclerosis (MS) is an autoimmune disease which affects myelin in the central nervous system (CNS) and leads to serious disability. Currently available treatments for MS mainly suppress the immune system. Regenerative medicine-based approaches attempt to increase myelin repair by targeting endogenous progenitors or transplanting stem cells or their derivatives. Fingolimod exerts anti-inf...

Isolation and in vitro Characterization of Mesenchymal Stem Cells Derived from the Pulp Tissue of Human Third Molar Tooth

Background: It is still controversial that the stem cells isolated from human dental pulp meets the criteria for mesenchymal stem cells (MSCs). The aim of the present study was to examine whether or not they are MSCs, or are distinct stem cells population residing in tooth pulp. Methods: Adherent fibroblastic cells in the culture of pulp tissue from human third molars were propagated through se...

Isolation, characterization and transduction of canine bone marrow-derived mesenchymal stem cells (cBM-MSCs)

BACKGROUND: Stem cell therapy in small animal medicineis still in its infancy and few in vitro and in vivo research projectsregarding animal Mesenchymal Stem Cells (MSCs) have beencarried out. On the other hand, Cell tracking is the first step of thecell-based therapies and is essential to recognize cell fate posttransplantation. OBJECTIVES: The aim of this study was toisolate, characterize, an...

Isolation, Characterization and Differentiation of Rat Adipose Tissue Derived Mesenchymal Stem Cells

Introduction:   Mesenchymal stem cells have the potential of self-renewal and differentiation into different cell types, including blood cells, heart, nerves and cartilage, and have unlimited power for division. These cells can be obtained from cord, before implantation from fertilized cells and also from various tissues of adults although the differentiation power and the ability to reproduce ...

Isolation, Culture, and Characterization of Human Dental Pulp Mesenchymal Stem Cells

Introduction Based on previous researches, dental pulp stem cells (DPSCs) are easily accessible with limited morbidity after collection. Their embryonic origin, from neural crests, explains their multipotency. DPSCs are primarily derived from the pulp tissues of the teeth. Objective: This study was undertaken to isolate, culture, and characterize two different third molar and first premolar hum...
